Hi everyone.

I'd like to know which way should I put the membrane during the blocking and washing step? Should I let the membrane floating on the solution or just let it lay on the bottom of the tank? or either way has nothing to do with the background? and what shaking speed is best for blocking and washing?

Put the membrane in the bottom of your box, with the buffer over the membrane. the protein should be on the top. Mix gently with the antibody, and faster with the washing buffer.
