CFDA-SE sample preservation

Hi!

I'm doing cell proliferation assay with CFDA-SE kit from Molecular-Probes. Because our FACS machine gets broken pretty often I have to fix my samples to preserve them. I've analysed 2-3 days old samples. Has anyone idea how long the fixed cells maintain proper signal when stained with CFDA?

I've gone out at least 2 weeks without any loss of signal. (Wrapped in foil and kept in the fridge)

What have you used to fix the cells? I'm using 2%PFA.

it honestly depends on what is in the fridge.  Usually between 2 to 4% PFA.  I fix with 100 uL of PFA for 20 minutes on ice and then fill the tubes to the top with Facs wash.  That dilutes out the PFA and provides enough liquid that my samples don't dry out.
