BSP primers

hi all,

I'm new to the filed of epigentics and a bit confused. I have a few basic queries.

1) what is a degenerate primer?
2) Is it necessary to include a CpG island in BSP primers?
3) I have the promoter sequence of the gene. Is it o.k, if i design BSP primers considering the sequence as the templete, irrespective of the fact that it is the sense or the antisense strand?

If anyone could help....it will great... :P

2). If you include a CpG in your primer sequence you are running no longer a BSP- but a MSP-PCR! BSP = Bisulfite Specific PCR, MSP = Methylation Specific PCR

3). It doesn't matter which strand you use for designing your primer, the methylation-information should be the same on both strands.
