1.why amido black can bind total transfer membrane protein?
2.ddH2O or ethanol,which one can add to the lower gel to prevent gas formation?

on gas formation just make your gel under a vaccuum then no bubbles

how to keep my gel in a vaccum condition?
i have water and alcohol only.
do i need any equipment?

are you making sds page gels ? triphasic? if so degas in a beaker with a vaccuum source attached then make sure the glass is TOTALLY clean and pour down one side of rig with pastuer pipette very slowly and carefully. it takes a little more time but it works for me.
