Every time I use PKH26 for the staining of MSCs I lose 90% of my cells. I use 2microM for the staining of 10millions of cells with final volume 2ml.

I was thinking to reduce the final concentration. Any idea?

We also had similar problem and we reduced the concentration of dye to half. Now it seems ok. BTW do you know the constituent of Diluent C ?
