As of late, my lab has been having a lot of difficulty transducing genes which are linked to kanamycin. We get very few colonies, and there are usually just as many on the control plate. We have tried using low-concentration Kn plates for the initial transduction and re-plating on high Kan, but if anything that only helps the control plates. It definitely wasn't always this way, just in the last 6 months or so...Anyone have any idea what might have happened? The transduction protocol has stayed the same, and still works with other antibiotics.
