Hi

I used to perform Trizol RNA extraction well. nice pellet good ration and good volume.  using fibroblast monolayer cell culture.
lately i started trying RNA extraction from chondrocytes and fibroblasts in high density 3D culture . but after precipitation with isopropyle i am getting gel like pellet. or as if it is two aquous phase one is very small amount around the pellet. i continued with RNA quantifiacation and as expected is not a good ration or volume
any idea why is this hapening . or what should i do ..
i got few normal white pellet in some samples.

thanks

i a, checking some troubleshooting and this problem seems to be called as liquid pellet!!!!

Hi,

You could try finding the constituent elements of the 3D matrix gel. What is it made of? That's probably the reason for the unusual precipitation. Can't really pin point the problem without knowing the above in advance.

Cheers

i am dealing with two kind of matrix

fibrin get
and alginate

i was expecting some problems with the alginate but not with the fibrin.
