Dear all,
I am having problems to produce a good titer of viral particules by using Phoenix amphopac cells.
A friend of mine told to select the cells with hygromycin and diphtheria toxin as the viral proteins and envelop are cloned in this cells in the same cluster of this resistance genes.
What do you guys thing abou it?
Is there another drug to substitute the diphtheria toxin as long as is a very difficult to find it?

Thanks,
Deivid

Hello :)

Yes, we made sure we selected our cells. Use 1ug/ml diphtheria toxin (Sigma) and 500ug/ml hygromycin (Sigma) for 2 weeks.
Hope this helps B)

Ok,
Thank you Clare.
Is necessary special care in the manipulation of the diphtheria toxin?
I mean special handling, wasting, etc.

Thanks again
