I transformed a gene in pichia about five years ago and the copy number using QPCR came as 5.2. Five years later and part of a stability study we performed a QPCR on a thawed vial of the cells and the copy number came at 10.8....What happened?? shouldn't the copy number be the same even after a hundred years??
