Hi!
I am extracting membrane protein (tagging and extracting) from cells and I am facing problem quantifying them. I use BCA and my standard curve is ok. I use 1mg/ml BSA in my respective buffer as my standard. and use diff concs. I also tried eluting my protein using less eluting buffer, but didnt help. I think BCA is not very sensitive in my case. I also made sure all components being used are compatible with the assay.
Please suggest help!!

You can't detect the protein by BCA or is it giving you variable readings?

Can you take a small aliquot and concentrate?
Do you have a "membrane" protein available to use as a control in the BCA?
