I use the following:
100mM 2-mercapto ethanol
2% SDS
62.5 mM Tris HCl PH6.7
100mM 2-mercapto ethanol
2% SDS
62.5 mM Tris HCl PH6.7

If it is PVDF membrane i use 0.2 N NaoH.
Its so convinient and time also less.
