how to break seed dormancy of arabidopsis

hi all

im trying to grow arabidopsis in solid agar media but the seeds donot grow .what can be other methods to make them grow ?
can anyone help?

What do you mean by solid agar media? is it 1/2MS containing agar? What is the germination %age of the seeds? How long do you keep the plates at 4 degrees for stratification?
I assume you are trying to grow mutants, right? You can grow some wild type seeds on the same media just to check in case if there is some problem with the media or with protocol.

Actually i had kept the arabisopsis seeds at 4C for 2 days and then i got germination of only a few arabidopsis seeds-4% very less regeneracy.
I am not certain whether keeping the seeds in constant motion is better than just keeping them in static condition at 4C.

Any suggestions

i suggest you to look for papers about breaking seed dormancy in arabidopsis, or to make a quick germination assay. good luck

perhaps the seeds were not stored properly...
In addition to stratefication treatment, you can treat seeds with 10 uM GA

another small question..is there any problem if I keep the seeds on plate in 4C bit longer time..like over 10 days and transfer them to culture room?? Would you expect any effect on germination rate??
