Hi!
I'm going to perform ion transport measurement in living cells by a fluorimetric method after loading cells with a fluorescent probe. I know that transport is ATP-dependent, but, after treating cultures with metabolic inhibitors (ouabain, azide, orthovanadate), I didn't observe the expected inhibition. I have the doubt that treatments are not efficacious. So, can you suggest me an alternative protocol to lower intracellular ATP levels (or enhance to conduct anyway a verification)? In that case, there a method not too complicated to verify if treatment is effective?
