How to calculate the insert and linear plsmid concentration for ligation
How much restriction enzymes need to be added for 1 micro gram of DNA IS it varies between plasmid DNA, and plant genomic DNA.
Please give me as much as possible explanation for these things.

Run your samples through a spectrophotometer and look at the absorbance at 260 and 280 nm - go to the protocols page on this site, under molecular biology there are a heap of links that will tell you all you need to know.
