Does anyone have a recipe for a "gentle" IgG elution buffer? I'm doing a Protein G magnetic bead purification of cell culture supernatant and I need an elution buffer that won't denature the protein G or antibody. I would buy a commercially available buffer, but I really want to get this done today! Thanks

You could try making a 0.1M glycine-HCl solution and keep the pH high (> 3). Also, be sure to add an alkaline buffer immediately afterward to neutralize the eluted fraction and minimize denaturation (Tris works).
