mean value of ct value

Hi all

please help me figure out this.

I have done many PCRs, but one of the reviewers of a paper want to know the MEAN VALUE OF CT (threshold cycle)

How do i calculate mean value of threshold cycle? do I use ct for
1) every plate?
2) every sample?
3) every gene?

I use gene of interest / reference gene method for normalising the value.

Hmm, do you mean real time pcr or normal pcr? Because I've never heard of having to predict ct for normal pcr (what is there to calculate anyway??? :lol: ).
For real time pcr you need this value for statistical significance. Have a look at these two sites:

Hi thanks for the reply . But my problem is not about how to determine the CT. My problem is what is meant by mean CT,? probably it is mean CT per gene I think. In that case I have to take individual values of each replicate CT and calculate the mean.
