I'm using j774 cell line for uptake studies.

My problem is that after 4/5 passages cells began to be spread and they didn't grow so fast as the beginning

could be a mycoplasma contamination?

J774 should grow like weeds......they adhere strongly to plastic and are very difficult to trypsinise. We grow them in suspension cultures using Techne Stirrer bottles.
If their morphology has changed then YES there could be some form of contamination. If they are not doubling every 15-18hrs then you have a serious problem.

Try and go back to a master bank......i.e. the lowest passage stock you have
