plasmid copy number

Hiiii

I want to determine plasmid copy number in one popular strain of E.coli. Currently I am using fluoresence method in corelation with cfu/ml of the culture. It is a tediously long procedure as well as accuracy is not so good. I found huge variation every time.Can anybody please tell me some quick and accurate method of determination of plasmid copy number?

Real-time PCR will be helpful in this context.  Maybe u can read the following journal:
