Anyone know how best to store viruses long term?

At -80C or in liquid nitrogen

no glycerol.
Just have them in what ever buffer they are in and freeze it in small aliquots. so you dont have to refreeze many times. But this also depends on the virus.

Is it ok to store the virus at -80 in the culture medium?

What buffer do you recommend? Is PBS ok?

a recommended buffer is SM buffer [from Sambrook]:
per liter:
NaCl = 5.8g
MgSO8 X 7 H2O = 2g
1M Tris-HCl (pH 7.5) = 50ml
2% (w/v) gelatin solution = 5ml
H2O to 1 liter

We keep in PBS or TEN buffer and store in -80.
