I have a question?

I am doing sputum processing and when I finhsed from the haemocytometer count, i have to centrifuge the the filtrate of the sample and then remove the supernatant and leaving the cell pellet, I need to know how THEN resuspend the cell pellet in PBS and then adjust the cells/volume to give me 0.5 million for preparation of cytospin slides!

Can anybody let me know whether 293T cells are attached (adherent) cells. Is there any need to coat the flask with Poly lysine for these cells. How many passage can be done with those.

The 293T (cultured in DMEM w/ FCS) I have used were adherent, but we used flasks that were already treated, so we didn't need to do anything to the flask. If culturing in different media you might have different levels of adhesion - for example using a serum free media seems to make them less attached and take on a different morphology.
