I am looking for a method to extract a protein from a coomassie/ silver stained gel.  I am then going to digest it with tryp so I need it as concentrated as possible

Try Elutrap, it is a realy convenient method for electroelution of proteins from SDS-PAGE. But it works better if you don't make a strong staining procedure (try amido black staining).

hai,
instead of amido black staining. u can use CuCl2.H2) (0.3M) for negative staining, cut the unstained bands and destain with 0.25M tris and 0.25M EDTA
