i am not able to find a protocol for gene knock out even after a through search,
please any one help me in this regard,i would be very thankful to them.

You'll have to give us more information. What organism do you want to knock a gene out of?

thank god there is some one to listen and help me out there.
first of all thank you for your reply"CLARE"
Ok now here i go
i have started working on this(gene knock out) from 4 days,i am just a beginner,so obviously my organism is E.coli the strain may be mostly DH5alpha and the knock out gene is dehydrogenase.
And i have found that lambda red recombinase method is used for knocking out the gene.
How would i get an good protocol so that i can start my work and do it uninterruptedly.
