Hi!

did anybody work with SfiI yet? Although I read the NEB manual I didn't get everything out of it. How does the enzyme work? If I flank my Insert by SfiI sites and clone it into my vector is it still like every other REN that the insert can be found in both orientations? or is the insertion different so that the insert will be inserted only in one orientation?

Hope you can fix my mind!
