Dear bioforumers
I am working in a project for construction of synthetic scfv,the Ab was tailored by the PI who is abroad now.
Please could anyone tell me how the design is made or suggest some readings or softwares.

I can send you some readings when I get into the lab.  What do you have for Ab now?  Is it a monoclonal?  If so, then the first step would be to sequence it, presumably by reduction, trypsin digestion and mass spec.  If it's polyclonal, you'll likely have to first make a monoclonal.  Once you have a protein sequence, then the conversion to an ScFv is reasonably straightforward, though not guaranteed to give good results.
