Hi All!

Has anyone recently tested the new Zymo CpG Methylase Kit?

CpG Methylase (M.SssI)

They seem to have another buffer and definately a different incubation temperature. Any experiences regarding complete methylation?

Hi All!

I give feedback on my own posting. I tested the Zymo CpG Methylase Kit after convincing my lab-leader that it should be worth to try. We had some problems receiving a complete methylation with the NEB M.SssI kit. With the Zymo kit methylation seems to be complete even for extremely CpG-rich regions for which the NEB kit had some difficulties. In all cases we weren't able to detect any residual unmethylated DNA.

The main differences are a different buffer and overnight-incubation at 30°C (NEB 37°C, 1-4h). Zymo ships their kit on dry-ice. Shipment by NEB is done on cool-packs. This seems to have a high impact on the stability especially of the SAM. This is the component we have identified to be most critical. Any comments on that?
