I did Southern blotting with human genomic DNA to know the gene copy number of my interesting gene. When I used my certain probe there were two signals for each lane, I used 3 kinds of restriction enzyme. Between these signals the density of those were different. I don't know the genomic sequence of my interesting gene, yet. Can anybody explain about the different signal? My probe is distributed differently to each digested genomic DNA because of the existence of intron? Actually I don't know the genomic sequence of my clone, I made the probe based on the cDNA sequence. Please help me...
