Hi,
has any one tried to express pGEX in the eucayotic cells or any comments?

I have many probems with pGEX4T1 vector too....
Now I think that BAD choice.... Have a others best vectors...
I need 2600 bp cloning insert with BamHI + SmaI digest and nothing at last!
My cells are eucaryotic too...
Best,
Beta.

The pGEX vectors are not designed or meant for protein expression in eukaryotic cells. They do not have a promoter for eukaryotes. They are designed for expression in prokaryotes and therefore have the correct sequences for prokaryotic transcription. These vectors are meant for expressing your protein in bacteria and will not work in eukaryotic cells.
