transformation of E. coli DNA in another E. coli host

Hello everybody,

I have a problem with the creation of a random whole genome library of one E. coli strain in one of the classical strains for transformation like DH10B and Top10. Have you ever seen any interference between two E. coli DNAs (studied and host ones) in a standard cloning procedure?

The transformation procedure seems OK (positive control) and both host strains are recombination and modification negative.

Any ideas?
