hello all,
i'm trying to extract keratin from chicken feathers.i'm really very much confused about the homogenization part.the extraction buffer contain 0.3M dtt.is it enough to add the extraction buffer diectly to the feathers and homogenate it or should i boil the feathers and add extraction buffer?is dtt alone enough to break the disulhifide bonds or should i boil the feathers to break the bonds before adding the extraction buffer to it?can anyone pls provide me with information.
