Hi,
I have problems in purification of my target protein using Qiagen Ni-NTA spin column under denaturing condition. there is an extra band found ard 47 kDa besides my target protein which is around 10 kDa. Can someone please help me? Thank you.

Use pH or imidizole gradient elution.
