I have a problem when transforming DH5alpha with a 6kb cDNA. The colonies, though turning out positive, will grow VERY slow and stay tiny. The bigger colonies on the plate are negative or tend to contain recombined cDNA . I never observe this problem with DNAs closely related to this one. Is there any clue whether I can enhance my positives with media or substitutes?? I use a very fat medium already (CircleGrow).
