Hi,

I have a question about side effects due to transfection prodcedures:

I transiently transfected Raji cells with a vector coding for a shRNA. Western Blot analysis revealed that my protein of interest seems to be affected by the electroporation as it decreases after transfection even in the controls.

I compared protein levels of untransfected cells with transfected cells (transfection with control and shRNA vector) on a western blot and my protein levels are in comparison to the untransfected cells significantly decreasing (the staining of actin is equal).
I would be very grateful if somebody would have an explanation for this or can give me paperlinks etc... .
