I will get the nuclear protein extract from cancer cells for EMSA.Almost all protocols suggest getting the cell pellet by scraping.Considering the time and work,I am thinking of typsinizing cells.Anyone ever did that before? Will it affect the quality of extracted protein? Thanks!

hi
i've never made a rigourous comparison of the two methods. But i would say that both methods are ok.
I choose the method regarding the number of plates i have.Till 20 plates, i scratch cells but more than 20 plates i prefer trypsin method that is quicker.

Obviously, scratching a 6 24 96 well-plate is quite impossible. :D
