southern hybridization temperature

hi,every professional,

   now i wanna pursue a low stringency southern hybridizaion. the probe is ~1kb and with nearly only 70% homology with the target DNA.
   how can i decide the hybridization temperature?

hi
as you get only 70%homology with your target, i would first try a room temperature assay...

but :) RT? can the double strand denature and accomplish hybridization successfully in such low temperature?

and do you mean i have to try a temperature grads?
