Antibiotics selection in mammalian expression

if my vector has an amphicilin and a neomycin selection markers. which one should i use for selection of my mammalian cells?

I am not sure... but my understanding is that the amp is for selection in bacteria when doing transformation and neo is for selection in mammalian cells when you want to establish stably expressing cells.

That's right. You should use ampicilling for selecting your vector on bacterial cultures, and neomycin/G418 when transfecting mammalian cells.
