I am trying to isolate dna from coffee leaves, which are brought from some other place and stored at -70C. But our efforts to isolate pure and good quality DNA have failed using both the SDS and CTAB method. The DNA we isolate seems to have lot of secondary metabolites in it and it is absolutely colourful and many timesdark in colour. We have tried mercaptoethanol, PVP and Na bisulphide, but none is satisfactory and we are presently purifying our DNA using column and then using it.
