HI Friends,
I am struggling with  a strange problem. I transformed pichia GS115 with pPICZalphaC having insert and selected the transforments on YPD-zeosin plate (100microgram/ml).

Few colonies appers on plate after incubation. But when I tried to sub-culture these colonies in liquid YPD media, I was shocked to see NO GROWTH. I have tried various concentration of ZEOSIN in liquid media but  results are same. I have also tried various concentration of Zeosin on YPD-zeosin plate and growth are fine even upto 300microgram/ml of zeosin.

Please help me if anybody have experinced this kind of problem earlier.
