Hi,
Please give me your opinions regarding expression cloning system for protease expression ( it has to be extracellular secretion), without affecting the host. Any idea about the use of Pichia expression system?

Are you interested in prokaryotic or eukaryotic expression system.
Expression in Bacillus subtilis results in secretion of the protein. You need to append suitable signal peptide ahead of you protein. Expression of such a construct in E.coli results in periplasmic accumulation.

Either one is ok, any idea about an eukaryotic system? The protease secreted must not affect the host itself.
