Hi,

I am working on cloning my gene using the pGEM-T easy kit. I already got the clone I want, checking from the EcoRI digestion.

However, I tried to extract the plasmid again and found out that the insert is gone. I only got the plasmid from the extraction.

I haven't heard anything about lossing the insert before....lossing of the plasmid would make more sense.

Anyone has any idea what has just happened in my culture?
