Hi my friends,
I got some problems in culturing PZ-HPV-7 cell line!
I've followed the  manufacture's instructions to culture PZ-HPV-7 cell line!
When I subculture it, it will attach!
But, it will die on the dish without deattach in another day!
Please help me to solve the problem and tell me what happend!

Hi,
I am growing PZ cell too. At the beginning, the cell didn't grow well. Then we use the trypsin inhibitor when we subculture it, the cell grows well. The media for PZ cell is KSFM. it is serum free, so the cell is sensitive to trypsin. I think that's the problem.

First of all, thank u for your replying.
There still something happend in my PZ cells, though I've used trypsin inhibitor to neutralize in subculturing step!
Besides, if I thaw another vial which are at  the same time to be freezed, PZ cells may grow happily after the first subculture.
Therefore, I always need to take a chance to culture PZ cells!
Is it usual in culturing PZ cells?

Actually, my PZ cells also have the same problem at first. Now it grows better, but the growth rate is still very slow. And the cell viability is low too. But if you keep the cell at high concentration when you subculture it, it may help. Never let the subcultivation ratio exceed 1:3.
By the way, what is the passage number of your PZ cell?

Wow, it sounds like PZ cells are not easy to thaw up at first! But if it is succeded to grow after 1st subculture, it will grow fast and enough to do research! Sometimes, the subcultivation ratio is 1:2, and I will keep up observating associated phenomena to find a good way for PZ growth.
Now, my PZ cells' passage is 35!
