Hi all,
i've gotten my RT PCR to work (Mg++ sucks big time), but i'm looking at my melting/dissociation curve... it's got one peak, but i'm wondering how do you know if the peak is at the right temp?  
i should be happy that there is just one, and it's nice, and pretty and... where abouts should this peak be?  I was thinking it sould be at about the same temp as required by the primers... or should it be higher or much higher....

should be happy its one peak, but have grown used to the fact that nothing really works well.... EVER.

going back to my corner now.

please help.
