Nuclear RNA knockdown with siRNA

Hello,

I'm trying to knockdown the amount of a nuclear RNA. Does the Dicer-mediated degradation of the targeted RNA occur in the nucleus or in the cytoplasm??

If it occurs in the cytoplasm, does polyadenylated nuclear RNA get exported to the cytoplasm before it is localized to the nucleas??

Is knocking down a nuclear RNA feasible?

There is a very recent paper reporting RNAi works in the nucleus. The abstract is here

my experience is that in the nucleus, antisense works a lot better than RNAi.

siRNAs can be delivered to the nucleus pretty efficiently with most cationic lipid-based reagents, and they induce cleavage of at least some nuclear-localized RNAs. Eg, Ambion launched recently siRNAs targeting non-coding RNAs, and I saw their data at the meeting - over 70% knockdown of several nuclear ncRNAs
