phospho-specific antibody

Hi again

Another problem. I'm using a phospho-specific antibody and I'm having problems. The antibody recognises another protein at the same size as the protein of interest. As a control I have a 18myc-tagged version of the protein. the phospho-antibody recognises the myc-tagged protein but also a protein at the same size as the wild type. I don't think that it is my protein of interest as a non-phospho antibody only recognises the myc-tagged version of the protein and not this mysterious X protein.

Although my problem may not sound very clear, does anyone have any ideas how I can remove this background band?

Try 2D gel. In theory, it should resolve any proteins.
