I'm having troubles making yeast protein extracts. When I use the TCA method my samples run as normal but I have problems with quantification of my samples. Any ideas about that?
However, whenever I try to use a Tris-based lysis buffer followed by bead beating the samples always run strangely. The bands are sometimes triangular in shape. What could this mean? I am really confused.
Can anyone help?
