Hi there! Has anyone ever worked on iNOS gene? I want to clone the iNOS gene .So for that I am isolating total RNA from rat liver tissue .By RT-PCR I tried to amplify the gene but was not successful though i changed the conditions and primers also but all in vain.Could anyone please suggest me something

Hi,
Is your RT reaction good? have you amplified other genes such as beta-actin from the RT? Does rat liver express iNOS? If not, find a source that expresses high levels of iNOS.
