I want to know what kind of method usually be used to prepare enough plasmid for transfection. I usually used the mini plamid preparation column of Qiagen from 5ml medium, but just can be used for several transfection. I want to know a method to prepare larger amount of plasmid at a time. Thank your attention!

hi,
i used the mtd. given in Maniatis lab manual. got a good yeild and no contaminating chromosomal DNA , very pure. its a bit long protocol but sure.

Just do maxpreps using a maxprep kit. You can grow bacteria in any volume of LB.
