Colleagues, could you share you approaches/experience in inserting 10xHis--TEV cassette upstream of coding sequence for a protein to be expressed (in yeast)?

Try out using the Ex-Site-Mutagenesis-Kit from Stratagene, if there is no targeting-sequence (for example for mitochondrial transit)
