Hi!!
Can I do an Klenow-Polymerase and following SAP-treatment in the same buffer as my restiction digest?!
Restriction digest buffer conditions: 20mM Tris (pH 8.5); 20mM MgCl; 1mM DTT; 100mM KCl.
I heard that Klenow-Pol works pretty much in any buffer, is that right?!
Happy science!!

Different enzyme needs different kind of circumstances.
you can find some information from NEB company.

Klenow and SAP will work in most restriction buffers.  When I cut vectors, I routinely perform the digestion in the presence of SAP.
