I have done "MSP" for about 3 months,but untill now I do not get any rusults.I use CHMICON's DNA modification Kit,and use Hotstart PCR.BUT my PCR has not nothing except primer dimer,my primer has worked by others.

Can you post your PCR conditions and your primer sequence?

Since you are using the chemicon kit, the problem may not be in the modification part. If you can get some working modified DNA from others, you can use that as positive control.

I performed Touchdown PCR:95-5m(1 cycle);95-40s,60-40s,72-40s(3 cycles);then annealing T is 58-40s(2 cycles),56-40s(2 cycles),54-40s(2 cycles),52-40s(2 cycles),50-40s(2 cycles),48-40s(30 cycles);finally 72-5m.
I now perform p16 and RASSF1A genes and the primers are all from the articles.
And in our lab,only me is performed "MSP".So it is more difficult to do this work.I really want to learn from you! THANKS!
