When we used pcDNA3.1 in DH5a, it was found that the vector was very difficult to be cut by EcoR I, I check the methylation information about the site, EcoR I wasn't sensitive to methylation. I don't the reason. Has some one met such kind of problem with the vector? which restriction enzyme work good in pcDNA3.1? Thank you in advance.
